Protein Sequencing General Information


In general, samples submitted for Edman sequencing should meet the following criteria. A single peak on HPLC or a clearly visible well separated band on polyvinylidene difluoride (PVDF) or equivalent blot, documentation of appropriate quantitation should accompany the sample. Synthetic peptides will be accepted in solution or lyophilized. See Sample Preparation for additional details.

The Protein Services Order Form can be downloaded in pdf format or picked up in 811 Vail. Please fill out the order form according to the instructions given. Completed order forms and your sample can be dropped off at the laboratory (811 Vail) during regular hours, Monday through Friday 8 am to 5 pm. If for some reason you can’t make it during these hours, call or email, we will make other arrangements. Be prepared to spend a little time talking to the laboratory staff about your sample. This is an important part of the protein sequencing process.

Price and Delivery Time: The order form lists current prices. Delivery time will depend on the number of samples queued up ahead of you. Ideally, a sample received on day one with a request for 20 amino acids can be put on the instrument the morning of day two, and the data can be obtained and analyzed by the morning of day three. For the anxious, some preliminary data may be available as the sequencing operation progresses. However, the interpretation of this data is subject to change when the complete data set is analyzed at the end of the run. You will receive a hard copy of your data set, a report with the sequence call, and a consultation with the operator to discuss the reliability of the data, problems or anomalies in the sequence.
