The Applied Biosystems Prism Model 7700 Sequencing Detection System is an automated PCR and fluorescence detection machine, which consists of an thermal cycler, laser, ccd module, a MAC G3 Computer that includes software for data collection and data analysis.
With the ABI Prism 7700 system, you can use real time PCR to quantitate DNA or RNA copy numbers. The SDS software collects and analyzes fluorescence generated during each PCR cycle.

To prepare your experimental DNA/RNA for real time PCR on the ABI Prism 7700, you can use standard laboratory methods for PCR.
Thermal Cycling and Detection
A fluorescent probe will specifically anneal to the DNA or cDNA and fluorescence is recorded following the 5' nuclease activity of the TAQ polymerase. Three specific annealing events must occur, therefore this system is highly specific and highly sensitive.
Real Time Data Observation
The raw data is analyzed one minute after the 2 hour thermal cycling is complete.
Data Analysis
At the conclusion of operation, the collected data is analyzed automatically. Raw data and analyzed data can be displayed on the computer screen. Data is processed by the software using a multicomponent analysis, baseline subtraction and scaling. A threshold cycle for each well is generated.
The sequence data can be exported to an Excel spreadsheet for further analysis.
