IV. Staining Formulas

A. Toluidine Blue

Add:

1 gram Toluidine Blue-0 stain
1 gram Sodium Borate
100 cc.double-distilled H2O

Dissolve completely (probable pH 12.0)


B. Uranyl Acetate (UA)

Add 4 grams uranyl acetate to 100 cc double distilled H2O, shake well and allow to dissolve for 48 hours. Use only the solution at least 1 cm above undissolved uranyl acetate.


C. Lead Citrate (Pb citrate)

In a clean 50 ml. volumetric flask add:

1.33 gm. Lead nitrate,Pb(N03)2
1.76 gm. Sodium Citrate, Na3(C 6H5O7 ) . 2 H2O

30 cc. CO2-free double-distilled H2O
Allow solids to dissolve for at least 30 minutes, shaking frequently
Add 8 cc.1N NaOH , shake until the solution is clear and add sufficient CO 2 -free double-distilled H2O to make 50 cc. Place in a brown bottle and maintain at room temperature, probable pH 12.0
