Scanning Electron Microscopy / X-Ray Microanalysis of Tissue Specimens

1. Obtain two unstained, mounted sections from block, and adjacent H&E stained section.

2. Remove paraffin in Toluene for 15 minutes, rinse in clean toluene, then and air dry one section. Rehydrate second unstained section (toluene through ETOH series to water).

3. Stain rehydrated de-waxed section with aqueous UA and Pb citrate. Stain for 5 minutes each in UA, and Pb citrate with thorough washing between stains. Air dry.

3. Carbon coat stained and unstained slides with fiber coater.

4. Analyze in the SEM with the aid of backscatter electron images and the X-ray microanalysis system. Use H&E slide as reference for areas of interest.

5. Record both secondary and backscattered images of areas of interest, as well as X-ray spectra of both background and specific items of interest. If needed, generate X-ray intensity maps, or line scans.
