A. 4% Glutaraldehyde with 0.1M Sodium cacodylate Buffer
Add: 1. 10 ampules (5 cc) each of 70% glutaraldehyde
2. 435 cc stock Na cacodylate solution (0.2M)
3. 385 cc double distilled H 2 O
Check pH and adjust to 7.2 to 7.5 if necessary with 0.1N NaOH or 0.1N HCl.
Check osmolarity, if not in range of 500 to 600 mOsm discard. Store at 4º C.
B. 8% Glutaraldehyde with 0.2M Sodium cacodylate Buffer
Add: 1. 2 ampules (5 cc each) 70% glutaraldehyde
2. 78.5 cc stock Na cacodylate solution
Check pH and adjust to 7.2 to 7.5 if necessary with 0.1N NaOH or 0.1N HCl.
Check osmolarity, if not in range of 1200 to 1400 mOsm, discard. Store at 4º C.
C. 4% Glutaraldehyde with 0.1 M Pipes Buffer
Add: 10 ampules (5 cc.) of 70% glutaraldehyde to
290 cc. 0.3M Pipes buffer stock,pH7.45 530 cc. double distilled H2O
Adjust pH to 7.35 with 1.0 N NaOH, mOsm=768,Store a 4º C.
