A. Stock Sodium cacodylate (Nacaco) 0.2M Buffer
DANGER: Sodium cacodylate is an arsenic compound and should never come in contact with the skin or be ingested.
Dissolve: 42.8 grams Sodium cacodylate in 1000 ml. in double distilled H 2 O
Adjust pH to 7.35 with 0.1N HCl
Store in refrigerator at 4º C
B. Sodium cacodylate buffer with 0.15M Sucrose
Add: 500 cc stock 0.2M Na cacodylate solution
500 cc double distilled H 2 O
52.85 gms. sucrose
Check pH and adjust to 7.2 to 7.5 with 0.1N NaOH or 0.1N HCl
Check osmolarity, if not in range of 325 to 340 mOsm, discard.
Store at 4º C
C. 0.3 M PIPES buffer stock
Dissolve: 45 grams Piperazine-N,N-bis-2-ethanol- sulfonic acid in
250 ml. double distilled H 2 O
Add: 13 cc. 12.5 N NaOH Adjust pH with 12.5 N NaOH to pH 7.35 Adding double distilled H 2 O make up to 500cc .
Osmolarity should be 713 mOsm
Store at 4º C
