TIP49 has WalkerA/WalkerB homology domains that
provide ATPase activity. Mutation of the WalkerB
motif (TIP49D302) creates an ATPase-deficient
protein that can still bind to TIP48 and c-Myc (A).
Oncogenic transformation by combined c-myc
and H-ras oncogenes is strongly suppressed by
cotransfection of the TIP49D302N mutant (B). The
TIP49D302N mutant does not suppress colony
formation with RSV-neo, indicating that the
protein is not inherently toxic to cells.
